Difference between locking and non-locking transmission

What is the difference between a locking and non-locking diff transmission? I found a replacement 5 spd for my car but it is a locking diff and I don't think my car has it. Will a locking diff go into a car that doesn't have it?

Yes it'll fit with a drivers' side axle change.
